similarities to normal histocompatibility antigens (4, 7, 8, 16, 24). Although the rejection of transplantable tumors among syngeneic animals has been used profitably in the study of animal tumors, this type of experimental design has been precluded in humans by both ethical considerations and the lack of a syngeneic donor-host relationship. Thus, a num ber of in vitro assays of the host's cellular and humoral immune responses against putative TSTA have been devel oped which are applicable to both animal and human tumor systems (1, 13, 14). Because the TSTA appear to be only weakly immunogenic, the antitumor immune responses evoked on the part of the tumor host are correspondingly weak (12, 23). Nevertheless, these immune responses are apparently far more tumor specific than can be evoked by the immunization of xenogeneic animals. On the basis of the foregoing observations, the expemi ments to be described were designed with the following objectives: (a) to use the host's antitumor immune response in order to monitor the purification of the putative TSTA of rodent and human tumor tissues; (b) to determine whether any structural and/or immunochemical similarities exist be tween human and rodent TSTA; and (c) to determine whether there are any immunochemical and/or structural relationships between the TSTA and normal histocompati bility antigens in either the rat or human.

Purification and evaluation of glycerol teichoic acid from biofilm forming Staphylococcus epidermidis as a putative vaccine candidate

Introduction: Biofilm forming Staphylococcus epidermidis is a main causative agent of infections related to medical devices. Purification and evaluation of Gly-TA polysaccharide from a biofilm-forming S. epidermidis as a putative vaccine candidate were the main goals of the current study. Methods: Taking advantage of size exclusion chromatography procedure, glycerol teichoic acid (Gly-TA) was p...

Virus oncogenesis and tumor immunogenicity in the mouse mammary tumor system.

Mammary tumorigenesis and mammary tumor trans plantation immunogenicity were examined in breeding fe males of 2 syngeneic sublines of the C3H strain: in mammary tumor virus (MTV) plus nodule-inducing virusinfected C3H/Sed mice; and in MTV-free, nodule-inducing virus-infected C3Hf/Sed mice. Ninety-seven % of the C3H mice (29 of 30) developed mammary tumors at an average age of 280 days. Of 43 di...

Expression Status of UBE2Q2 in Colorectal Primary Tumors and Cell Lines

Background: Activation of the ubiquitin-proteasome pathway in various malignancies, including colorectal cancer, is established. This pathway mediates the degradation of damaged proteins and regulates growth and stress response. The novel human gene, UBE2Q2, with a putative ubiquitin-conjugating enzyme activity, is reported to be overexpressed in some malignancies. We sought to investigate the ...

Generation of antigenic variants from a nonantigenic murine tumor cell line by transfection with a gene encoding a novel tumor-specific transplantation antigen.

Tumors induced in mice by UV radiation often express highly immunogenic tumor-specific transplantation antigens (TSTA). The 216 gene, which encodes a TSTA of the C3H tumor UV-1591, has been cloned and characterized as a novel major histocompatibility complex Class I antigen. The purpose of this study was to determine whether the 216 gene-encoded TSTA can function as a tumor rejection antigen wh...

Functionality of androgen receptor-based gene expression imaging in hormone refractory prostate cancer.

PURPOSE A highly augmented, prostate-specific two-step transcriptional amplification (TSTA) method was developed with the ultimate goal of delivering an effective and safe gene-based treatment to prostate cancer patients. Because very limited treatment options are available for recurrent hormone refractory prostate cancer (HRPC), it is imperative to assess whether the prostate-specific antigen ...
